چکیده

Abstract Current treatments have shown limited success in reversing DM induced vascular endothelial barrier dysfunction. Vascular endothelium forms the inner most lining of blood vessels, regulates variety biological processes such as angiogenesis, wound healing and host defense mechanisms. During inflammatory conditions, diabetes mellitus myocardial infarction cell-cell junctions start to disrupt because internalization junctional proteins, (VE) cadherin. This leads formation minute inter-endothelial gaps, infiltration protein-rich fluid immune cells interstitial space. If remains unchecked, persistent buildup edema underlying sets stage for serious life-threatening complications. Therefore, we hypothesized that donepezil prevents high glucose-induced dysfunction by preventing destabilization proteins. We investigated potential protective role Donepezil glucose increase human coronary artery (HCAE) permeability process. Wound assay was performed employing electric signals both monitor process monolayer. To investigate signaling mechanism involved protection offered Donepezil, found prevented loss VE-cadherin junctions, abrogated stress fiber cell exposed high-glucose solution. In conclusion, our study shows maintained function improved glucose. Chicago State University CTRE grant Professor Nadeem Fazal, MD, PhD

P 61: MicroRNA as a Therapeutic Tool to Prevent Blood Brain Barrier Dysfunction in Neuroinflammation

Endothelial cells present in brain are unique and differ from other peripheral tissues in a number of ways, which ensures specific brain endothelial barrier properties. Endothelial dysfunction is the earliest event in the initiation of vascular damage caused by inflammation. Various microRNAs (miRNA) have been discovered in different cellular components of the blood bran barrier (BBB). miRNAs a...

Arginase inhibition improves endothelial function in patients with coronary artery disease and type 2 diabetes mellitus.

BACKGROUND Endothelial dysfunction plays an important role in the early development of atherosclerosis and vascular complications in type 2 diabetes mellitus. Increased expression and activity of arginase, metabolizing the nitric oxide substrate l-arginine, may result in reduced production of nitric oxide and thereby endothelial dysfunction. We hypothesized that inhibition of arginase activity ...

The Therapeutic Effects and Role of MiRNAs in Coronary Artery Disease: A Systemic Review

Cardiovascular diseases are the most common causes of human morbidity and mortality despite significant therapeutic improvements by surgical or pharmacological approaches in the last decade. MicroRNAs (miRNAs) have a significant role in diseases development or response to treatment. In this review, we summarized the role of miRNAs in the pathogenesis or the treatment of coronary artery disease ...

Coronary artery disease and diabetes mellitus.

Diabetes mellitus (DM) is a major risk factor for cardiovascular disease. Near-normal glycemic control does not reduce cardiovascular events. For many patients with 1- or 2-vessel coronary artery disease, there is little benefit from any revascularization procedure over optimal medical therapy. For multivessel coronary disease, randomized trials demonstrated the superiority of coronary artery b...
